Description Stat3 is activated by a variety of cytokines including interleukines, CNTF, and EPO (1,2). Activation of Stat3 is accompanied by tyrosine phosphorylation at Tyr705 which induces dimerization, nuclear translocation, and DNA binding (1–3). Transcriptional activation also appears to be regulated by serine phosphorylation (Ser727) probably via MAP-like kinases (4,5) (Figure 1). As phosphorylation of Stat3 at Tyr705 is essential for dimerization and DNA binding, phosphorylation at this site is an excellent marker of Stat3 activity (1–3). The Stat3 Antibody Kit provides reagents and protocols for the rapid analysis of Stat3 phosphorylation status. The Kit includes a phosphoantibody that recognizes Stat3 only when phosphorylated at Tyr705, a control Stat3 antibody (phosphorylation-state independent), protein controls and protein markers for western blots, and the HRP Western Detection System.
