Recombinant Mouse Wnt-3a induces beta -catenin responsive Topflash activation in HEK293T cells. The specific Wnt pathway inhibitor, endo -IWR 1, inhibits Topflash activity from a 500 ng/mL dose of Recombinant Mouse Wnt-3a with a typical ED 50 < 0.2 μM. The less active diasteriomer exo -IWR 1 Wnt inhibitor shows minor inhibitory activity in this assay. Recombinant Mouse Wnt-3a induces alkaline phosphatase production in the MC3T3-E1 mouse pre-osteoblast cell line. The ED 50 for this effect is ≤ 5 ng/mL. The specific Wnt pathway inhibitor, endo -IWR 1, inhibits alkaline phosphatase production induced from a 10 ng/mL dose of Recombinant Mouse Wnt-3a with a typical ED 50 < 0.8 μM. The less active diasteriomer exo -IWR 1 Wnt inhibitor shows a > 5 μM ED 50 with a Recombinant Mouse Wnt-3a dose of 10 ng/mL.
