| Bioactivity | Binds STAM protein with Kd = ~7 µM. In in vitro assays containing 200 nM AMSH, a ~10-fold increase in the rate of hydrolysis of K63-linked diubiquitin FRET substrate was achieved upon the addition of STAM. Half-maximal increase occurred at 0.4 µM STAM. Typical enzyme concentration for use in vitro ranges from 100-500 nM depending on conditions and substrate. Suggested Reaction Buffer for plate-based assays is 50 mM Hepes, pH 8.0, 100 mM NaCl, and 0.1 mg/ml BSA. AMSH activity is partially or fully inhibited by DTT at concentrations above 1 mM. |
