SEE ALSO...

GRK 3 Antibody (C-11) is an IgG1 κ mouse monoclonal GRK 3 antibody (also designated GRK3 antibody, BARK2 antibody, or ADRBK2 antibody) that detects the GRK 3 protein of mouse, rat and human origin by WB, IP, IF and ELISA. GRK 3 Antibody (C-11) is available as both the non-conjugated anti-GRK 3 antibody form, as well as multiple conjugated forms of anti-GRK 3 antibody, including agarose, HRP, PE, FITC and multiple Alexa Fluor® conjugates. Heterotrimeric G protein-mediated signal transduction is a dynamically regulated process with the intensity of signal decreasing over time despite the continued presence of the agonist. This phenomenon, referred to as agonist-mediated desensitization, involves phosphorylation of the receptor by two classes of enzymes. The first class is comprised of the second messenger-regulated kinases, such as c-AMP dependent protein kinase A and protein kinase C. The second class includes the G protein-coupled receptor kinases (GRKs). At least seven members of the GRK family have been identified. These include rhodopsin kinase (GRK 1), two forms of β-adrenergic receptor kinase: GRK 2 (βARK, βARK1) and GRK 3 (βARK2), IT-11 (GRK 4), GRK 5, GRK 6 and GRK 7. Phosphorylation of receptors by GRKs appears to be strictly dependent on the receptor being in its agonist-activated state.

Our HEK GRK3 KO cell display a band for GRK3 when probed for this Ab, even using the recommended light chain secondary. The same non-specific signal is seen with fluorescence. I have also followed the recommended protocol. Is there any additional advice?

What is the recommended fixation method for immunofluorescence staining with GRK 3 (C-11): sc-365197 monoclonal antibody?

Asked by: Dr Ninau Qelp
Thank you for your inquiry. We recommend fixing cells for 5 minutes in -10°C methanol and allowing to air dry. The full protocol can be viewed here: https://www.scbt.com/scbt/resources/protocols/immunofluorescence-cell-staining

Rated 5 out of 5 by from Excellent antibody for Western blotting. This product has been independently tested and validated in-house at Kinexus Bioinformatics Corporation as a very potent and highly specific antibody for Western blotting with a mixture of mouse brain, testes and thymus tissue lysates. In immunoblots, it very strongly detects an ~70 kDa protein, which is slightly lower than the predicted size of about 80 kDa.
