Smo Antibody (E-5) is an IgG2a κ mouse monoclonal Smo antibody (also designated Smoothened antibody) that detects the Smo protein of mouse, rat and human origin by WB, IP, IF and ELISA. Smo Antibody (E-5) is available as both the non-conjugated anti-Smo antibody form, as well as multiple conjugated forms of anti-Smo antibody, including agarose, HRP, PE, FITC and multiple Alexa Fluor® conjugates. Overexpression of either Wnt-1 or the GLI proteins results in cancer; however, the molecular basis for this transformation was poorly understood. The Wnt-1 and GLI proteins have now been placed in a signaling cascade downstream of the mammalian homologs of the Drosophila hedgehog and patched proteins. The Drosophila segment polarity gene hedgehog (hh) encodes a secreted protein that appears to function in embryonic and imaginal disc patterning. The ptc gene, also identified as a Drosophila segment polarity gene, encodes the transmembrane protein patched, the expression of which is precisely regulated during embryonic development. Hedgehog has been shown to enhance the expression of the Wnt family of proteins through a signaling cascade involving the GLI transcription factors, while patched functions as a repessor opposing the effects of the hedgehog. Smoothened (Smo), a seven transmembrane receptor, is complexed with patched in many tissues and is believed to be an essential component in the Hh signaling pathway.

Hello, my lab works with S. purpuratus sea urchins. Could you please check if the antibody will be compatible with our species? This is the NCBI reference sequence for the Smo protein in urchins: XP_030830155.1

Has this antibody been tested in Xenopus, either WB or IF?

Thank you for your question. Anti-Smo Antibody (E-5) has only been tested for detection of Smo of mouse, rat and human origin by WB, IP, IF and ELISA. It has not been tested in samples from Xenopus.

For Western Blot, is it recommended to use denatured or non-denatured conditions with Smo Antibody (E-5): sc-166685?

Rated 3 out of 5 by from good for western blot analysisWe used this antibody to detect Smo and got a good result. Fresh liver samples were homogenized in RIPA buffer, then 40ug of total protein were subjected to 10% gel for SDS-PAGE and transferred onto a PVDF membrane. After blocking with 5% skim milk in 1×TBS-T, membrane were incubated overnight at 4℃ with Smo antibodies(1:1000).And then, it were incubated with anti- mouse IgG(1:2000) at room temperature and detected with ECL kit.
