Lipoxygenases are a family of enzymes which dioxygenate unsaturated fatty acids, thus initiating lipoperoxidation of membranes and synthesis of signaling molecules, as well as inducing structural and metabolic changes in the cell. The Lox enzymes in mammals include 12-LO and 15-LO, which are classified with respect to their positional specificity of the deoxygenation of their most common substrate, arachidonic acid. The metabolism of arachidonic acid leads to the generation of biologically active metabolites that have been implicated in cell growth and proliferation, as well as survival and apoptosis. 15-Lipoxygenase (15-LO) acts in physiological membrane remodeling and the pathogenesis of atherosclerosis, inflammation, and carcinogenesis. It is highly regulated and expressed in a tissue- and cell-type-specific fashion. IL-4 and IL-13 play important roles in transactivating the 15-LO gene. Overexpression of 15-LO type 1 in prostate cancer contributes to the cancer progression by regulating IGF-1R expression and activation.

I would like to use this antibody to perform flow cytometry on both mouse and human samples. I can't find any references showing it's use in flow cytometry, does it work for this application?

I have a question. Does this antibody react to both Alox12 (platelet type 12-LO) and Alox15 (white blood cell type 12-LO (12/15-LO)) in mice? What is the recommended dilution ratio when staining with frozen sections?

Hi, It says that this antibody detect platelet and leukocyte 12 lo. Does that mean that it detect by IF ALOX 15 and ALOX12

I'd like to use this antibody to detect the protein 15-LO, in other words, 15-LOX-1. So I wonder if this antibody can also be able to detect 15-LO2 which would be bad for our specific research. Thanks!

Thank you for your question. This mouse monoclonal antibody is specific for detection of 15-LO and is not expected to cross-react with 15-LO2. We have not experimentally validated this, but based on the epitope the antibody is specific for 15-LO.

I see a doublet above the predicted 75kDa weight and some other bands above 100kDa and above 150kDa. Not sure if the doublet bands above 75kda are the right ones.
