Aldolase C Antibody (H-11) is an IgG1 κ mouse monoclonal Aldolase C antibody (also designated ALDOC antibody, ALDC antibody, Fructose-bisphosphate aldolase C antibody, or Brain-type aldolase antibody) that detects the Aldolase C protein of mouse, rat and human origin by WB, IP, IF, IHC(P) and ELISA. Aldolase C Antibody (H-11) is available as both the non-conjugated anti-Aldolase C antibody form, as well as multiple conjugated forms of anti-Aldolase C antibody, including agarose, HRP, PE, FITC and multiple Alexa Fluor® conjugates. Fructose 1,6-bisphosphate aldolase catalyses the reversible condensation of glycerone-P and glyceraldehyde 3-phosphate into fructose 1,6-bisphosphate. Fructose 1,6-bisphosphate aldolase exists as three forms, the muscle-specific Aldolase A, the liver-specific aldolase B, and the brain-specific aldolase C. Aldolase A, B, and C arose from a common ancestral gene, from which aldolase B first diverged. Aldolase A is one of the most highly conserved enzymes known, with only about 2% of the residues changing per 100 million years. Aldolase B is regulated by the hormones insulin and glucagon and has been implicated in hereditary fructose intolerance disease (3,5). Aldolase C is a polypeptide that is exclusively expressed in Purkinje cells (6,8-10). Aldolase C-positive Purkinje cells are organized in the cerebellum as stripes or bands that run from anterior to posterior across the cerebellum and alternate with bands of Aldolase C-negative Purkinje cells (7,10).

'Is treatment with Triton X-100 needed when using this antibody for immunofluorescence cell staining?' Thank you for your question. You can find our recommended IF protocol on our website as Protocol 5: Immunofluorescence Cell Staining. If fixing in formalin, we recommend you include a cell permeabilization treatment. After fixing the cells, wash in three changes of PBS. Then incubate for 3 minutes in a solution of 0.01% Triton X-100 in PBS. If you have further questions, feel free to contact our technical service directly.

Is this antibody very specific to Aldolase C? Can it detect other isoforms of aldolase, such as aldolase A (I want to distinguish these two isoforms)?

Rated 1 out of 5 by from Does not work wellThe new batch of Aldolase C antibody (H11) gives very high background when used at prescribed 1:200 dilution. It does not work at 1:500 dilution. It does not work as well as it should. We should ask about a fresh batch of the antibody.
