Bax Antibody (B-9) is an IgG2b κ mouse monoclonal Bax antibody (also designated BCL2 associated X antibody or BCL2L4 antibody) that detects the Bax protein of mouse, rat and human origin by WB, IP, IF, IHC(P) and FCM. Bax Antibody (B-9) is available as both the non-conjugated anti-Bax antibody form, as well as multiple conjugated forms of anti-Bax antibody, including agarose, PE, FITC and multiple Alexa Fluor® conjugates. The Bax protein both homodimerizes and forms heterodimers with Bcl-2. Although Bax has extensive amino acid homology with a related protein designated Bcl-2, the Bax Antibody (B-9) specifically detects Bax without showing cross reactivity with Bcl-2. Overexpression of Bax accelerates apoptotic death induced by cytokine deprivation in an IL-3 dependent cell line and Bax also counters the death repressor activity of Bcl-2. The Bcl-2 gene was isolated at the chromosomal breakpoint of t-bearing follicular B cell lymphomas. Bcl-2 blocks cell death following a variety of stimuli and confers a death-sparing effect to certain hematopoietic cell lines following growth factor withdrawal. Bcl-2 is localized to outer mitochondrial membranes and endoplasmic reticulum as well as nuclear membranes.

 I run my sample on 10 % gel, I used 1/1000 in 5% BSA in TBS with  3h incubation, and I didn’t get any bands. Would you please advice me. 1)  What is the recommended dilution? The  dilute should be milk or BSA?  2) How long should I Incubate?What gel %?

Thank you for your question. We would recommend using the starting dilution for Western blot (1:200) and blocking your membrane with, and incubating the antibodies in 5% nonfat milk in TBST. I have three questions: 1) What gel percentage do you use to separate Baxα and Baxβ? 2) Does Bax Antibody (B-9): sc-7480 works for IF like the (6A7) clone: sc-23959? 3) If so, in human, in mouse cells or both species' cell lines?

Thank you for your question. We are running a 15% SDS-PAGE gel for detection of Bax with this antibody. Both antibodies are recommended for IF on mouse, rat and human samples.

can i use Bax Antibody (B-9) in goat? if yes what procedure do i follow?

Thank you for the question. We recommended Bax Antibody (B-9) for detection of Baxα and Baxβ of mouse, rat and human origin by WB, IP, IF, IHC(P) and FCM. Unfortunately, we have not evaluated this antibody on samples derived from other animal species. Therefore, we are unable to predict cross-reactivity with the Bax protein from goat.
