CD3-ζ Antibody (6B10.2) is an IgG1 κ mouse monoclonal CD3-zeta antibody (also designated CD247 antibody, or CD3 zeta antibody) that detects the CD3-zeta protein of mouse, rat and human origin by WB, IP, IF, IHC(P) and FCM. CD3-ζ Antibody (6B10.2) is available as both the non-conjugated anti-CD3-zeta antibody form, as well as multiple conjugated forms of anti-CD3-zeta antibody, including agarose, HRP, PE, FITC and multiple Alexa Fluor® conjugates. The T cell antigen receptor (TCR) recognizes foreign antigens and translates such recognition events into intracellular signals that elicit a change in the cell from a dormant to an activated state. Much of this signaling process can be attributed to a multisubunit complex of proteins that associates directly with the TCR. This complex has been designated CD3 (cluster of differentiation 3). It is composed of five invariant polypeptide chains that associate to form three dimers: a heterodimer of gamma and epsilon chains (γε), a heterodimer of delta and epsilon chains (δε) and a homodimer of two zeta chains (ζζ) or a heterodimer of zeta and eta chains (ζη). The ζ and η chains are encoded by the same gene but differ in their carboxyl-terminal ends due to an alternative splicing event. The γ, ε and δ chains each contain a single copy of a conserved immunoreceptor tyrosine-based activation motif (ITAM). In contrast, the ζ chain contains three consecutive copies of the same motif. Phosphorylated ITAMs act as docking sites for protein kinases such as ZAP-70 and Syk and are also capable of regulating their kinase activity. The crystal structure of the ZAP-70 SH2 domains bound to the ζ chain ITAMs has been solved.

In the description "raised against amino acids 36-54 mapping within an N-terminal domain of CD3-ζ of human origin" However, aa 36-54 is almost in the TM domain (31-51). How do you define aa 36-54? with or without leader? can it detect cytoplasmic one

Thank you for your question. The exact epitope recognized by this mouse monoclonal antibody has not yet been determined. We do have IHC(P) data on our website using this antibody to detect membrane and cytoplasmic staining in formalin-fixed paraffin-embedded tissue sections.

Is the agarose-conjugated CD3-ζ Antibody (6B10.2): sc-1239 provided as 500µg/mL, unlike the other formats that come as 200µg/mL? The recommended concentration for IP is 1-2µg/mL--does this mean that you would only need 2-4µL agarose-conj. Ab per 1mL IP?

Thank you for your question. For the agarose conjugated antibody, sc-1239 AC, we recommend using ~ 20uL of a 25% slurry, which equals 10 ug of ab/5 uL of agarose beads. Each vial contains 500 ug of antibody/250 uL of agarose in 1 mL of slurry (PBS + Sodium Azide + conjugated antibodies).

What is the recommended dilution for IHC/P application?

Thank you for your question. The recommended starting dilution for IHC(P) as described in the antibody’s datasheet is 1:50 with a dilution range of 1:50-1:500.

I am using the LiCor/Odyssey system. Which secondary antibody do you recommend using with CD3-ζ Antibody (6B10.2): sc-1239?

We recommend using one of our our exclusive Mouse IgG Binding Proteins (m-IgGκ) conjugated to CFL 680 or CFL 790. They are suitable for use with Near-Infrared (NIR) detection systems, such as LI-COR/Odyssey and other comparable systems. We recommend using a directly conjugated primary antibody. This product, sc-1239, is available conjugated to FITC, AF647, and AF488.
