C3 Antibody (B-9) is an IgG1 κ mouse monoclonal C3 antibody (also designated C3 antibody or Complement C3 antibody) that detects C3 precursor, C3a anaphylatoxin, C3 α chain, C3 β chain and C3b α′ chain of mouse, rat and human origin by WB, IP, IF, IHC(P) and ELISA. C3 Antibody (B-9) is available as the non-conjugated anti-C3 antibody form, as well as multiple conjugated forms of anti-C3 antibody, including agarose, HRP, PE, FITC and multiple Alexa Fluor® conjugates. Complement C3 precursor contains complement C3 beta chain, complement C3 alpha chain, C3a anaphylatoxin, complement C3b alpha chain, complement C3c fragment, complement C3dg fragment, complement C3g fragment, complement C3d fragment and complement C3f fragment. C3a, C4a, and C5a are potent anaphylatoxins that are released during complement activation, a system of ligand-surface protein interactions specific to cells of hematopoietic lineage that aids in the elimination of pathogens. C3a and C5a secretion correlates with pathophysiological phenotypes such as asthma and bacterial meningitis. Binding of these proteins to their respective G protein-coupled receptors (C3aR, C5aR), which are present on the surface of myeloid leukocytes, induces proinflammatory events such as cellular degranulation, smooth muscle contraction, arachidonic acid metabolism, cytokine release, leukocyte activation and cellular chemotaxis. C3aR is expressed in brain and activated B-lymphocytes whereas C5aR is prevalent on the surface of hepatocyte, lung, smooth muscle, and endothelial cells. Upon activation, C3aR and C5aR are susceptible to rapid GRK-mediated phosphorylation and clathrin-coated vesicle targeting. C5aR utilizes the Ras-Raf-ERK1/2 cascade and couples to Gi/G16 proteins.

hello,i see C3 antibody (B-9): sc-28294 is a mouse monoclonal antibody,how can it detect c3a anaphylatoxin and C3b α′ chain at the same time,if i want to use it in IHC,how can i distinguish it is C3b,C3a or c3?

Thank you for your question. Yes, C3 Antibody (B-9): sc-28294 is recommended for detection of C3a anaphylatoxin in mouse sample by WB, IP, IF, IHC(P) and ELISA. We don't have an in-house data for detection of C3a anaphylatoxin in mouse sample by WB at this moment, but please refer to the citations for this product.

Can I use non-transfected HEK-293T as a negative control? In your legend of Near-infrared western blot analysis of C3 expression said 293T has C3 band. But same time in your "sc-28294 HRP. Direct western blot analysis" legend said 293T has no band.

Thank you for your question. Our Control 293T Lysate: sc-117752 is derived from non-transfected 293T cells and suitable for use as a negative control with our 293T Transfected Lysates. Our Control 293T Lysate goes through the same transfection process as our gene-specific transfected lysates, except without transfecting any cDNA into the cells. The Control 293T Lysate is also used at a lower concentration than regular cell lines for Western blotting. That said, it does appear that HEK293 does not have significant RNA expression of C3 per the Human Protein Atlas. We will review of the Near-infrared Western blot data showing a signal in HEK293T cell lysate, and if it can not be supported we will remove it.

Has anyone used this AB for IHC in rat brain tissue?

Thank you for your question. We do not have publications specifically in rat brain tissue, but we have many publication in other rat tissues such as kidney, intesting and heart. We do have publications in mouse brain samples, so I would expect it to work perfectly fine in rat brain as well.

Hi - I see that this antibody can detect C3 (which has a molecular weight of 150kDa), but that it is also recommended to detect C3a and C3b. How do I specifically probe for C3a and C3b separately on a western blot? Do I add in something to break C3 down?

Thank you for your question. At this time, we are not able to provide any specific inductions or treatments for samples to increase expression of the different C3 subunits. This is a research-based question, and we would recommend you consult with the primary literature. There are many product citations using this antibody for Western blotting applications, so it may be useful to start by reviewing the citations on our website for this antibody.

Under what conditions have you demonstrated a positive C3 signal in human paraffin samples? I have tried numerous epitope retrieval protocols and seen nothing. I'm disappointed that you discontinued the goat polyclonal V-20 in favor of this product.

We use a 1:50 dilution with this antibody on paraffin sections, after a antigen retrieval in sodium citrate with heat. Please contact our Technical Service Department by phone, e-mail or live chat. How can C3 Antibody (B-9): sc-28294 be used for double or triple staining, if the other primary antibodies are also raised in mouse?

In this case, we recommend using a directly conjugated mouse monoclonal primary antibody. This mouse monoclonal antibody is currently available conjugated to phycoerythrin (sc-28294 PE), fluorescein (sc-28294 FITC), Alexa Fluor® 488 (sc-28294 AF488) or Alexa Fluor® 647 (sc-28294 AF647).

Rated 1 out of 5 by from Does not detect human C3 in paraffin tissuesI tried using this antibody to detect human C3 in paraffin-embedded tissues, using three different antigen retrieval techniques, and saw nothing. I also tried using the unconjugated antibody followed by an anti-mouse secondary, saw nothing. On the other hand, the discontinued goat polyclonal worked.
