SEE ALSO...

Fibronectin Antibody (EP5) is an IgG1 κ mouse monoclonal Fibronectin antibody (also designated FN1 antibody) that detects the Fibronectin protein of mouse, rat and human origin by WB, IP, IF, IHC(P) and ELISA. Fibronectin Antibody (EP5) is available as both the non-conjugated anti-Fibronectin antibody form, as well as multiple conjugated forms of anti-Fibronectin antibody, including agarose, HRP, PE, FITC and multiple Alexa Fluor® conjugates. Fibronectin is an extracellular matrix glycoprotein present on most cell surfaces, in extracellular fluids and in plasma. A high molecular weight heterodimeric protein, it was originally discovered as a protein missing from the surfaces of virus-transformed cells, and it has been shown to be involved in various functions including cell adhesion, cell motility and wound healing. Alternative splicing and glycosylation give rise to several different forms of Fibronectin, some of which exhibit restricted tissue distribution or association with malignancies. It has been shown that Myofibroblast phenotype formation correlates with the occurrence of glycosylated Fibronectin and Fibronectin splice variants in Dupuytren′s disease.

Can anyone share a working protocol for immunostaining for sc-8422 AF488 for mouse tissue?

Asked by: shihan
Thank you for your question. Please refer to our IF protocol for direct staining (using a directly conjugated primary antibody, without any secondary antibody for detection). https://www.scbt.com/scbt/resources/protocols/immunofluorescence-cell-staining

This antibody (EP-5) is suggested as a replacement for the polyclonal N-20 antibody. Where is the epitope for EP-5? Will it recognize the 70 kDa N-terminal fragment of human fibronectin?

Asked by: KlaraB
Thank you for your question. The exact epitope recognized by this mouse monoclonal antibody has not yet been determined. We have not specifically evaluated this antibody for reactivity with the 70 kDa N-terminal fragment of human fibronectin, but based on the Western blot data on our website, we have not observed any 70 kDa fragment specifically in the samples we have tested.

Is it possible to use the Fibronectin Antibody (EP5) for immunohistochemistry applications if I am working with mice tissue samples?

Asked by: Trav11
Yes, this is absolutely possible. We recommend using our directly conjugated mouse monoclonal primary antibodies for instances like this. Currently, the Fibronectin Antibody (EP5): sc-8422 is available conjugated to HRP (sc-8422 HRP), phycoerythrin (sc-8422 PE), fluorescein (sc-8422 FITC), Alexa Fluor® 488 (sc-8422 AF488) or Alexa Fluor® 647 (sc-8422 AF647).
