In this experiment, apoptosis was induced in Jurkat and 143B osteosarcoma cells by FAS and also by treatment with staurosporine (HeLa cells were also treated, but only with STS). Mitochondrial and cytoplasmic fractions were isolated (using kit Cell Fractionation Kit ab109719/MS861) and probed using ab110415 (MSA12). As is clear from the gels, cytochrome c has translocated partially in FAS-induced cells and STS-treated osteosarcoma cells, and almost completely in STS-treated Jurkat and HeLa cells. The three control targets allow for verification of the "cleanness" of the cell fractionation.

5x10e7 Jurkat cells were cultured in the absence (1-2) or presence of 2 uM Camptothecin (CPT) (ab120115) for 24 hours (3-4) or with 10 uM CPT for 4 hours (5-6). 30 uL cytosolic (1, 3, 5) and mitochondrial (2, 4, 6) extracts were loaded onto the gel. Membranes were probed with anti-Cytochrome C Mouse MAb (ab6311) (dilution 1:200) followed by Goat Anti-Mouse IgG (HRP) (ab97040) (dilution 1:2000).Bands were detected at the prediced size of 12 kDa.

Two dimension Blue Native PAGE analysis of fibroblasts that are (A) normal and (B) complex I deficient. It is clear that the complex I deficient cell line shown in B shows no detectable level of complex I. Results demonstrate the formation of ubiquitin thioester linked E1 and/or E2 conjugates in all +Mg-ATP reactions. The absence of such conjugates in negative control reactions demonstrates that their formation is Mg-ATP dependent (required for E1 activation) and hence derived from the ubiquitin cascade.
